MADS-box家族PpCMB1基因调控花发育分子机制

摘 要:

花器官的发育是重要的农艺性状, 因此候选基因的筛选十分重要。MADS-box基因家族在调控花发 育中发挥了重要作用, 在这篇论文中, 对桃(Prunus persica) MADS-box家族PpCMB1 (PpMADS2)基因进行 了研究。从春雪桃花中克隆了PpCMB1基因, 通过定量PCR进行组织特异性分析, 结果显示该基因在雌 蕊与萼片中表达, 这表明PpCMB1与桃花的发育密切相关。将PpCMB1转入拟南芥(Arabidopsis thaliana),  与野生型对照相比, PpCMB1过表达株系具有显著促进抽薹早花的表型。通过酵母双杂交(Y2H)实验与双 分子荧光互补(BiFC)实验证明PpCMB1PpDAM5蛋白互作, 提示PpCMB1蛋白可能通过抑制PpDAM5白的功能促进花发育。本研究为调控桃花发育的基因研究提供了新的见解。

Molecular mechanism of PpCMB1 gene in peach MADS-box family  regulating flower development

Abstract:

The development of floral organs is an important agronomic trait, so the selection of candidate  genes is very important. The MADS-box gene family plays an important role in regulating flower development. In this paper, the PpCMB1 (PpMADS2) gene of the MADS-box family in peach (Prunus persica) is  studied. The PpCMB1 gene was cloned from the flowers of ‘Chunxue’ peach, and tissue-specific analysis by  quantitative PCR showed that the gene was expressed in pistils and sepals, this indicates that PpCMB1 is  closely related to the development of peach blossoms. PpCMB1 was transferred to Arabidopsis thaliana. Compared with the wild-type, PpCMB1 overexpression lines had a significant phenotype of promoting early bolting.  The yeast two-hybrid experiment and BiFC assay proved that PpCMB1 can interact with PpDAM5 protein, which  indicates that PpCMB1 protein may promote flower development by inhibiting the function of PpDAM5 protein.  This study provides new insights for the study of genes regulating peach flower development.
